RESUMO

The experimental research was carried out on the juvenile carp (Cyprinus carpio L.). The impact from supplemental feeds consisting of variable concentrations of chelate compounds, biogenic trace elements and probiotic lactobacillus-based product Bacillus subtilis VKPM B-2335 was evaluated. Optical and qualitative parameters of the lactobacillus base were studied in order to identify the major group of substances potentially able to influence the end result. The purpose of this research was to identify changes in the structure of the zymogen granules and their dimensions at which supplemental feeds produce a stimulating effect on the synthesis of zymogens in exogenous cells of the secretory part of pancreas. At the outcome of the study, for the first time, it was possible to prove that the integrated action of chelates and lactobacillus-based probiotics complemented each other. Metal chelate compounds contributed to enlargement of the zymogen granules, if compared to the control values. The bacterial products accelerated production of the zymogen granules in acinar cells diffusely located in carp hepatopancreas.


A pesquisa experimental foi realizada em carpas juvenis (Cyprinus carpio L.). O impacto de suplementos alimentares consistindo em concentrações variáveis de compostos quelatos, oligoelementos biogênicos e produto probiótico baseado em lactobacilos Bacillus subtilis VKPM B-2335 foi avaliado. Parâmetros ópticos e qualitativos da base de lactobacilos foram estudados a fim de identificar o maior grupo de substâncias potencialmente capazes de influenciar o resultado final. O objetivo dessa pesquisa foi identificar mudanças na estrutura dos grânulos de zimogênio e suas dimensões nas quais alimentos suplementares produzem um efeito estimulante na síntese de zimogênios em células exógenas da parte secretora do pâncreas. No resultado do estudo, pela primeira vez, foi possível comprovar que a ação integrada dos quelatos e dos probióticos à base de lactobacilos se complementava. Compostos quelatos metálicos contribuíram para o aumento dos grânulos de zimogênio, se comparados aos valores de controle. Os produtos bacterianos aceleraram a produção dos grânulos de zimogênio nas células acinares localizadas difusamente no hepatopâncreas da carpa.

ObjectiveTo understand the concentration of heavy metal cadmium and cadmium in portunus and mantis shrimp, and to timely identify food safety problems and potential hazards. MethodsPortunus and mantis shrimp samples from different provinces were collected and categorized based on different regions and locations, and some samples were made from tissue parts. Graphite furnace atomic absorption spectrometry was used to detect cadmium items, the cadmium exposure of portunus and mantis shrimp was evaluated simultaneously ResultsThe detection rate of cadmium in 124 batches of portunus sold in Shanghai was 100% (124/124), the detection rate of cadmium in 63 batches of mantis shrimp sold in the market was also 100% (63/63). The cadmium content varied in different tissue parts, and the cadmium enrichment in hepatopancreas was the highest in the edible parts of portunus and mantis shrimp. The average detection value, 50th percentile value, 95th percentile value of cadmium in the hepatopancreas of portunus accounted for 52.64%, 49.28% and 98.65% of the PTMI, respectively. The average detection value, 50th percentile value and 95th percentile value of cadmium in the hepatopancreas of mantis shrimp accounted for 30.76%, 32.04% and 46.16% of the PTMI, respectively. ConclusionThe average residual levels of heavy metal cadmium in portunus and mantis shrimp are within the safe range.

This work aimed to assess the effect of the probiotic strain, Lactobacillus plantarum, on the levels of leptin, IGF-1 and their receptors on the hepatopancreatic tissues of Nile tilapia (Oreochromis niloticus) and then correlate fish growth performance and gut microbiological parameters. Fish juveniles (±23g) were reared in a recirculation system with constant aeration and temperature (25°C). They were distributed into six polyethylene tanks (45L) and fed twice a day at 5% of the tank biomass with the respective diets: control (commercial diet without probiotic) and supplemented with L. plantarum inoculum (1 x 108 CFU mL-1), both in triplicate. After 30 days of feeding, L. plantarum-fed fishes showed greater weekly growth rate, final weight, and feed conversion rate, in addition to higher count of lactic-acid bacteria and lower count of pathogenic bacteria in the intestinal tract, when compared to the control group. The immunostaining intensity for IGF-1 and leptin hormones was lower after L. plantarum supplementation than in the control group, with no change in the level for receptors. This reduction could implicate important changes in fish metabolism and homeostasis.(AU)


O presente trabalho teve como objetivo avaliar o efeito da cepa probiótica Lactobacillus plantarum sobre os níveis de leptina, IGF-1 e seus receptores no tecido hepatopancreático de tilápia-do-nilo (Oreochromis niloticus) e correlacionar com o desempenho zootécnico e os parâmetros microbiológicos intestinais dos peixes. Juvenis de tilápia-do-nilo (±23g) foram distribuídos em seis tanques de polietileno (45L) conectados a um sistema de recirculação, com aeração e temperatura constantes (25°C). Os peixes foram alimentados duas vezes ao dia, a 5% da biomassa do tanque, com as respectivas dietas: controle (dieta comercial sem probiótico) e suplementada com L. plantarum (1 x 108 UFC mL-1), ambas em triplicata. Após 30 dias de cultivo, os peixes alimentados com L. plantarum apresentaram maiores ganho de peso semanal, peso final e conversão alimentar, bem como maior contagem de bactérias ácido-láticas e menor contagem de bactérias patogênicas no trato intestinal das tilápias alimentadas com dieta probiótica, em comparação ao grupo controle. A intensidade da imunomarcação para os hormônios IGF-1 e leptina foi menor com a suplementação de L. plantarum do que no grupo controle, sem alterar os níveis de seus receptores. Essa redução pode implicar mudanças importantes no metabolismo e na homeostase dos peixes.(AU)

Abstract Study aimed to evaluate the effect of replacing fish meal with different levels of full fat soybean meal (FFSBM) on growth and digestive enzyme activities in the stomach, Liver and intestine for Clarias gariepinus. Four diets (D1, D2, D3 and D4) were formulated with 0, 15, 20 and 20 g 100-1 protein + DL-methionine by alternating FFSBM with fish meal. The growth of C. gariepinus was found to be significantly decreased when FFSBM replacement increased. Final body weight was 89.69, 79.70, 70.82 and 68.29 g for fish fed on D1, D2, D3 and D4, respectively, with significant differences between treatments. Specific growth rate (SGR) ranged between 3.11 to 2.78%. Proteolytic activity was higher only with alkaline pHs, whereas only very low activity was shown with acidic. Results of liver showed approximately similar results at acid and alkaline. In contrast, higher proteolytic activity in the stomach was observed at acid pHs 3.0 and 4.0 μg tyrosine-1 minute-1 mg-1 protein) whereas lower values were observed at neutral pH 7.0 g tyrosine-1 minute-1 mg-1 protein for catfish fed on the experimental diets. Moreover, trypsin activity was higher for the stomach, followed by the intestine and liver. However, higher amount of amylase observed in the liver than intestine and stomach.


Resumo Objetivou-se avaliar o efeito da substituição da farinha de peixe por diferentes níveis de farelo de soja integral (FSI) sobre o crescimento e atividades de enzimas digestivas no estômago, fígado e intestino de Clarias gariepinus. Quatro dietas (D1, D2, D3 e D4) foram formuladas com 0, 15, 20 e 20 g 100-1 proteína + DL-metionina, alternando FSI com farinha de peixe. O crescimento de C. gariepinus foi significativamente reduzido quando aumentou a substituição de farinha de peixe por FSI . O peso corporal final foi de 89,69, 79,70, 70,82 e 68,29 g para peixes alimentados com D1, D2, D3 e D4, respectivamente, com diferenças significativas entre os tratamentos. Taxa de crescimento específico (TCE) variou entre 3,11 a 2,78%. A atividade proteolítica foi maior apenas com pHs alcalinos, enquanto que atividades muito baixas foram somente mostradas com ácido. Os resultados no fígado mostraram valores aproximadamente semelhantes tanto em meio ácido quanto alcalino. Em contraste, observou-se maior atividade proteolítica no estômago em pH ácido (3,0 e 4,0 g de tirosina-1 minuto-1 mg-1 de proteína), ao passo que valores mais baixos foram observados em pH neutro (7,0 g de tirosina-1 minuto-1 mg-1 de proteína) para os bagres que se alimentaram com as dietas experimentais. Além disso, a atividade da tripsina foi maior para o estômago, seguida pelo intestino e pelo fígado. No entanto, observou-se maior quantidade de amilase no fígado,intestino e estômago.

Gambusia holbrooki es el pez de agua dulce con mayor distribución en Chile y el mundo, pero los estudios que abordan la morfología e histología hepática del pez son escasos. El hígado es utilizado para evidenciar efectos subletales de contaminantes ambientales y es preciso contar con una descripción histomorfológica del hígado para futuros estudios comparativos. El presente estudio tiene como objetivo describir patrones histomorfológicos del hígado de Gambusia holbrooki, para ello se colectaron 97 individuos adultos de los sistemas límnicos de los valles de Lluta y Azapa (Extremo norte de Chile), para observar patrones morfológicos comunes en ambas poblaciones, se utilizó técnicas histológicas de rutina e histoquímica. Las evidencias demostraron que el hígado contiene tejido pancreático y su arquitectura tisular es trabecular con mayor presencia de capilares sinusoides.


Gambusia holbrooki is the freshwater fish with the greatest distribution in Chile and the world. However, studies dealing with morphology and liver histology of fish are scarce. The liver commonly shows the sublethal effects of environmental pollutants and there should be a histomorphological description of the liver for further comparative studies. The present study aims to describe histomorphological patterns of the liver of Gambusia holbrooki. A total of 97 adult individual specimens were collected from the ecosystems in the valleys of Lluta and Azapa (Region of Arica and Parinacota), to observe patterns morphologically common in both populations. Routine histological and histochemical techniques were used for analysis. The evidence showed that the liver contains pancreatic tissue, and that tissue architecture is trabecular with greater presence of capillary sinusoids.

Aim: To determine acute toxicity (96 hrs) of malachite green for Lamellidens marginalis and to assess it’s biochemical consequences and antioxidant response against control. Methodology: Fresh water bivalve, Lamellidens marginalis were collected from Rajaram tank, Kolhapur and acclimatized for 7 days. To ascertain the acute effect (96 hrs) of malachite green dye, static bioassay was conducted at 0.3, 0.4, 0.5, 0.6, 0.7, 0.8, 0.9, 1.0, 1.1, 1.2 ppm on fresh water bivalve, Lamellidens marginalis. The rate of mortality was recorded and LC0 and LC50 values for 96 hrs were calculated. Estimation of biomolecules like protein, glycogen and lipid and antioxidant activity in different tissues of the organism was estimated. Results: The LC0 and LC50 values were determined by static bioassay method. The effects of LC0 and LC50 concentrations of MG were evaluated by examining the biochemical profile and antioxidant response against control in different tissues like gill, mantle, hepatopancreas and gonad. The maximum significant decrease in glycogen (-29%) (-40.86) and protein (-31.73%) (-43.63%) content was observed in gills at both the concentrations LC0 and LC50 respectively as compared to other tissues. There was highly significant depletion in protein content of all the tissues. Lipid content in all tissues at LC0 concentration showed moderately significant depletion (p <0.01), while at LC50 concentration highly significant depletion (p< 0.001) was recorded. From SOD, CAT and GPx enzymes, SOD enzyme activity increased significantly (p < 0.001) in all tissues at LC50 concentration. Gill and heptopancreas showed significant increase in antioxidant enzymes response at both, LC0 and LC50 groups. Interpretation: Malachite dye induces toxicity, by lowering biochemical content and inducing antioxidant enzyme activity.

Commercially important alkaline phosphatase can be recovered from the hepatopancreatic tissue of Indian white shrimp with optimum yield, activity, and purity by carefully designing the criterions of ion exchange chromatographic purification. The tissues were homogenized, clarified, and concentrated. Concentrated homogenate was purified with optimum yield, activity and purity by optimising criterions of chromatography such as pH and ionic strength of the binding buffer, and ionic gradient and the flow rate of the elution buffer in DEAE-cellulose column. The enzyme was optimally bound to the column at pH 8.4 and ionic strength 0.1 M NaCl at flow rate of 1mL/min, and eluted with high resolution at ionic gradient of 0.10-0.35 M NaCl in 25 min at a flow rate of 1.5mL/min, and hence is the optimum criterions of chromatographic purification.

Objective. The present study aimed to investigate the presence of vibrios in the hepatopancreas of cultured shrimp. Materials and methods. Vibrios from the hepatopancreas of fifteen samples of five specimens each, of apparently healthy Pacific white shrimp (Litopenaeus vannamei) were isolated, identified and quantified. Results. The vibrio density ranged from 430 to 2,400 MPN g-1 (rs MPN cm-1=-0.114; rs MPN g-1 = 0.211). Thirty isolations were obtained, most of which belonged to the species V. cholerae (n=11) and V. parahaemolyticus (n=7). Conclusions. The outcomes of the present study suggest that, even in the absence of symptoms of vibriosis, the microbiota of the hepatopancreas of cultured shrimp may include sucrose positive and negative vibrios.


Objetivo. El presente estudio tuvo como objetivo investigar la presencia de vibrios en el hepatopáncreas del camarón de cultivo. Material y métodos. En este estudio, fueron aislados, identificados y cuantificados los vibrios del hepatopáncreas de 75 camarones blancos del Pacífico (Litopenaeus vannamei), aparentemente sanos, oriundos de un cultivo en la región nordeste de Brasil. Quince muestras, cada una consta de cinco camarones, se pusieron a prueba. Resultados. La densidad de Vibrio varió de 430 a 2.400 NMP g-1 (rs NMP cm-1 = -0.114; rs NMP g-1 = 0.211). Treinta aislamientos fueron obtenidos, la mayoría de los cuales pertenecían a la especie V. cholerae (n=11) y V. parahaemolyticus (n=7). Conclusiones. Los hallazgos del presente estudio sugieren que, incluso en ausencia de síntomas de la vibriosis, la microbiota de lo hepatopáncreas del camarón de cultivo puede incluir vibrios sacarosa positivos y negativos.

This study aimed at determining the level of thiaminase in the Limicolaria flammea with a view to ascertaining the nutritional and health implications of its consumption. Methodology: Proximate analyses of two tissues, headfoot and hepatopancreas, were investigated to assess their nutritional qualities. The thiaminases in the tissues were purified by chromatographic separations using DEAE-Sephacel ion - exchange and Biogel P-100 columns. Results: The Michealis Menten constant obtained for hepatopancreas thiaminase was 0.83 mM and 0.13 mM for thiamine and aniline respectively; for the headfoot thiaminase, 1.06 mM and 0.16 mM was obtained for thiamine and aniline respectively. An optimum pH of 8.5 was obtained for thiaminase in the two tissues. Temperature optimum of 65°C and 70°C was obtained for the headfoot and hepatopancreas enzymes respectively. The amino acids and glutathione activated the enzyme from hepatopancreas, while the headfoot enzyme was significantly inhibited except proline which showed very high activation. The cations: NH4 +, Hg2+, Mn2+, Na+, Ni2+, and Zn2+ tested showed no inhibition of the enzymes in both tissues. Increased concentrations of 2-mercaptoethanol, 6-amino hexanoic acid and ethylenediaminetetraacetic acid (EDTA) inhibited the activities of thiaminases from the two tissues. Conclusion: The study concluded that the tissues, hepatopancreas and headfoot of Limicolaria flammea contained high level of thiaminase. This may have medical implication in its consumption as a good source of high quality protein.

Gafrarium divaricatum were exposed to xylene (4.25 and 8.50 mg l-1), benzene (4.35 and 8.70 mg l-1) and gear oil-WSF (1 and 2%) for 30 days. Chronic exposure of clams to the pollutants resulted in loss of bubbling epithelium, reduction in cytoplasm volume and density, fusion of cell membranes and nuclei forming darkly stained area at basal part of the cells. Disintegration of basement membrane due to damaged epithelial cells, disruption of inner lining of tubule, formation of necrotic spaces, separation of epithelial cells from basement membrane, increase in internal luminar area, complete necrosis of epithelial cells as well as occurrence of cell debris in between the tissue were also observed in the clams due to chronic exposure of the toxicants.

The present work describes the effect of different salinities on the functional morphology of the P. argentinus hepatopancreas and analyses the tissue recovery after re-acclimation to freshwater. Adult prawns of both sexes at sexual rest were collected from a tributary of the Mar Chiquita coastal lagoon. The prawns were acclimated in aquaria to four salinity conditions: 0 (control), 8, 16 and 24‰. To evaluate the possible tissular recovery, after 60 days individuals from all the treatments were gradually acclimated to freshwater and maintained for other 30 days. Hepatopancreas samples were processed at the beginning of the trial and every 30 days using standard histological techniques for OM and TEM. The individuals from all the treatments, except the controls, showed a continuous weight decrease, and survival was lower when higher the salinity. At 30 days from the beginning of the experiment, hepatopancreas from 16 and 24‰ salinities showed an enlarged tubular lumen and an infolded basal lamina. Ultratructurally, nuclear retraction, cytoplasmolysis, and RER membranes separated with electron-dense content were observed in all the treatments except 0‰. After 60 days, profound alterations were observed with the three treatments. After the re-acclimation period, there was no reestablishment of the functional cytology. The tolerance to short-term salinity changes explains the capability of this prawn to inhabit in estuarine environments.

The juveniles of M. malcolmsonii were exposed to 24.1 µg l-1 of Hg for a period of 21 days. The gills and hepatopancreas of test prawns were sampled and processed for electron microscopic observations. Mitochondria are the organelle most affected in the gills of test prawns. The number of mitochondria and the electron-density of the matrix were found to be less in test prawns. The in-folding of cell membrane associated with mitochondria was absent in test prawns. This suggests that operation of the mitochondrial pumps was affected in the gills of test prawns. Vacuoles with crystalline granular inclusions were noted in the gills of test prawns. These are suggestive of metal-rich inorganic deposits or granules representing detoxified dumps of Hg. In the hepatopancreas of test prawns, the tubules exhibit vacuoles with granular inclusion and the cell cytoplasm contains electron-dense granules, which indicate a storage detoxification of Hg. The mitochondria were shrunken in the hepatopancreas of test prawns. This suggests attenuation of its function. The rough endoplasmic reticulum appeared vesiculated and dilated. These reactions denote the hyperactivity of the rough endoplasmic reticulum. Membranous whorl-like structures with myelin fibers and residual bodies were seen in the hepatopancreas of test prawns. Such structures indicate the involvement of lysosomal breakdown in detoxification process. The ultrastructural alterations are suggestive of the operation of compensatory mechanisms within the test prawns to enable it to tolerate Hg toxicity. However, these alterations would have an impact on the cellular integrity of the gills and hepatopancreas and such alterations can be taken as ‘biomarkers’ for assessing Hg pollution in the aquatic environment.

The present study examined alterations in the protein pattern and histopathology of the hepatopancreas and gills of Macrobrachium malcolmsonii following exposure to Sevin®, the commercial grade of carbaryl (1-Naphthylmethyl carbamate) pesticide. Juvenile prawns were exposed to sub-lethal concentrations 5.15 'g l-1, 7.73 'g l -1and 15.47 'g l -1of carbaryl for 21 days. Samples were obtained from the hepatopancreas and gills of prawns. The concentration of buffer soluble protein in tissues of test prawns was significantly (p<2.5%) lower than the control. This suggests that proteins were utilized to generate energy in order to withstand carbaryl induced toxic stress. Polypeptide bands of different molecular mass (150 - 10 kDa) in tissues of test prawns stained less intensely than those in control. This indicates that carbaryl toxicity caused protein degradation in tissues of test prawns. The histology of the hepatopancreas and gills showed few marked pathological changes in prawns exposed to carbaryl. These include rupture of the basal laminae, abnormal infiltration / aggregation of hemocytes in the interstitial sinus of the hepatopancreas and in the hemocoelic space of gill lamellae, fused gill lamellae and necrosis like appearance both in the tubules of the hepatopancreas and gill lamellae. The structural alterations observed in the hepatopancreas and gills of the prawns suggest that carbaryl caused tissue damage in M. malcolmsonii. Protein degradation is a phenomenon in damaging tissues, and thus, under such condition the hepatopancreas and gills of test prawns would not perform their vital functions, which in turn ultimately affect the survival and growth of M. malcolmsonii. The changes noted in protein and histology of test prawns can be taken as biomarkers for monitoring pesticide pollution in aquatic environments. Since this species of prawn is a nutritious delicacy to human being, monitoring aquatic pollution by carbaryl is warranted.

In decapod crustaceans, the digestive gland is concerned with the digestion, absorption of nutrients, the storage of reserves and excretion. The metabolism and the histological and histochemical changes of the hepatopancreas are observed in response to physiological demands as moult, reproduction, digestive process. Thus the hepatopancreas structure should be recognized to provide important morphological information to future studies involving the nutrition requirements of freshwater prawn culture. In this study, second-generation Macrobrachium amazonicum produced from wild broodstock collected in the state of Para in Brazil were used. Thirty adult male and female M. amazonicum were selected and randomly transferred to five experimental units for macroscopic and microscopic studies. The hepatopancreas of M. amazonicum is a large, yellowish-brown, compact organ, which occupies much of the cephalothoracic cavity. It has right and left halves that are enclosed together in a laminar connective tissue capsule, and at the same time they are separated by an interstitial connective tissue. The two halves are thereby called the right and left hepatopancreatic lobes. The principal tubule gives rise to four secondary tubules at each hepatopancreatic lobe. The morphological and functional unit consists of a blind-ended hepatopancreatic tubule, considered in the present study as the hepatopancreatic lobule. Each hepatopancreatic tubule can be subdivided into distal, medial and proximal zones. The hepatopancreatic tubule is lined by a pseudostratified epithelium that consists of five different cell types, which include the E-cell (embryonic), F-cell (fibrillar), B-cell (blister-like), R-cell (resorptive) and M-cell (midgut or basal). It is important to emphasize that the function of each cell type in the hepatopancreas during the digestive cycle is not yet established for decapods.


La glándula digestiva en los crustáceos decápodos asume las funciones de digestión, absorción de nutrientes, almacenamiento de las reservas energéticas y excreción de metabolitos. El metabolismo y las alteraciones histológicas e histoquímicas son observados como respuesta a necesidades fisiológicas, tales como: muda, reproducción y procesos digestivos. Por lo tanto, se requiere conocer la estructura histológica del hepatopancreas con el fin de reunir información morfológica para futuros estudios que consideren las necesidades nutricionales para el cultivo de los camarones de agua dulce. Fueran utilizados 30 animales, machos y hembras de Macrobrachium amazonicum, producidos a partir de reproductores colectados en el Estado de Para, Brasil. Las observaciones macro y microscópicas permitieron concluir que el hepatopancreas del M. amazonicum es un órgano grande y compacto de color amarillo tendiendo a marrón, que ocupa la mayor parte de la cavidad céfalo-torácica. El órgano presenta dos mitades, derecha y izquierda, las cuales están envueltas por una cápsula de tejido conjuntivo, y al mismo tiempo, separadas por tejido conectivo intersticial. Las dos mitades del órgano son identificadas como lobos derecho e izquierdo. Cada lobo presenta un túbulo principal que origina cuatro túbulos secundarios. Por lo tanto, la unidad morfofuncional del hepatopancreas consiste en un túbulo hepatopancreático de fondo ciego, considerado en esta investigación como lóbulo hepatopancreático. Cada lóbulo hepatopancreático puede ser dividido en 3 regiones: distal, media y proximal, y en su totalidad, se observa revestido por epitelio seudo estratificado que reúne cinco tipos celulares: célula E (embrionaria), célula F (fibrilar), célula B (globosa), célula R (reabsortiva) y célula M (basal). Es importante enfatizar que la función de cada tipo celular del hepatopancreas no está todavía aclarada para los decápodos.

We analyzed the morphological and functional state of hepatopancreas in Palaemonetes argentinusfrom two environments with different pesticide concentrations. Los Padres lagoon (Argentina) is an area subjectedto contamination due to the slow exchange of water, the shallow depth and the input of contaminatedwater. Prawns living in this lagoon accumulate high amounts of organochlorine pesticides in their tissues. Hepatopancreas of prawns from Canal 5, an adjacent shallow stream where the amount of pesticides is belowtoxic levels, and from Los Padres lagoon were processed by standard histological techniques with light microscopyand transmission electronic microscopy. At Los Padres lagoon, we found important tissular alterations,such as intertubular infiltration of haemocytes and connective tissue, epithelial retraction in some tubules, anda folded basal lamina. Important necrotic desquamation, with cariolysis, cariorrexis and lack of cellular detailswere also observed. Numerous tubules presented an enlarged and irregular lumen with the epithelium atrophiedor completely absent. In general, the lesions were particularly located in the medullar region of the organ. Atthe ultrastructural level, R and F cells were the most damaged. Both cell types had nuclear retraction, chromatincondensation and cytoplasmic lysis. Some R cells also had dilated mitochondria and numerous lysosomes, and the basal cytoplasm was nearly completely lysed. The hepatopancreas of prawns from Canal 5 did not evidenceany alterations. The histopathological study of the hepatopancreas is a highly sensitive tool to evaluate thephysiological condition of prawns and water quality. Other environmental conditions were similar, so it can beassumed that pollutants were the main cause of organ deterioration.


Analizamos el estado morfológico y funcional del hepatopáncreas de Palaemonetes argentinus de dosambientes con diferentes concentraciones de plaguicidas. La laguna Los Padres (Argentina) es un área sujeta a contaminación debido al lento intercambio del agua, la escasa profundidad y el influjo de agua contaminada. Las gambas acumulan aquí grandes cantidades de plaguicidasorganoclorados en sus tejidos. Los hepatopáncreas de gambas del Canal 5 y de la laguna Los Padres fueronprocesados mediante técnicas histológicas estándar para microscopía óptica y electrónica de transmisión. Loshepatopáncreas de los individuos recolectados en Los Padres tenían alteraciones tisulares importantes, comoinfiltración intertubular de hemocitos y tejido conectivo, retracción epitelial en algunos túbulos y láminas basalesplegadas. También se observó descamación necrótica importante, con cariolisis, cariorrexis y falta de detallescelulares. Vimos muchos túbulos con un lúmen irregular y agrandado, con el epitelio atrofiado o completamenteausente. En general las lesiones se localizaron en la región medular del órgano. Ambos tipos celulares mostraronretracción del núcleo, condensación de la cromatina y ruptura del citoplasma. Algunas células R también teníanmitocondrias y numerosos lisosomas dilatados, y el citoplasma basal casi completamente desintegrado. Nohallamos alteraciones en los hepatopáncreas de gambas deCanal 5. El estudio histopatológico del hepatopáncreas es una herramienta muy apropiada para evaluar la condiciónfisiológica de las gambas y la calidad del agua. Por la semejanza de otras condiciones ambientales, suponemosque los contaminantes fueron la causa principal del deterioro de los órganos.

Objective After an experimental exposure to low dosage of Cd 2+(39*!?mol/L) for 30 days,the changes of Cd 2+ on ultrastructure of the hepatopancreas cells of the freshwater crab (Sinopotamon yangtsekiense♂)at three different times were studied. Methods Sixty freshwater crabs(♂)were divided into two groups:control(n=30) and test(n=30)groups.The test(n=30) group was exposed to low dosage of Cd 2+(39*!?mol/L)while the control crabs were in water without cadmium.During the 5*!d,15*!d,30*!d,the ultrastructure of the hepatopancreas cells was observed by using transmission electro-microscopy(TEM). Results In the course of the experiment,the nuclear membrane changed from spreading all over the place to disintegrated,and the penetration of the mitochondrial outer membrane and inner membrane was changed.The mitochondria showed swelling and vacuolating.Mitochondrial cristae partly disintegrated,then fully disintegrated.Besides,the Cd 2+ exposure affected the gross form and the structure of the rough endoplasmic reticulum(RER).At first,the RER became the different sized vesicles,then,the ribosomes on the little vesicles began to fall,and finally,the RER turned to the concentric round lamellar structures.But the smooth endoplasmic reticulum(SER)increased markedly.In addition,the numbers and types of lysosome were increased in the mean time and some lysosomes became void vesicles,the autophagosomes or the myeloid bodies(MD).The microvilli showed partly falling and vacuolating.Conclusion After the cadmium treatment the detoxification mechanism of the freshwater crab's hepatopancreas cells lost,the structural and functional integrality of the mostly organelles,such as the nucleous,the mitochondria and the rough endoplasmic reticulum,were destroyed,so that the normal physiological function of hepatopancreas cells was affected.

The regional distribution and relative frequency of some endocrine cells in the pancreas of the carp, Cyprinus carpio Linnaeus, belonging to the family Cyprinidae in the order Cypriniformes, were observed using specific mammalian antisera against insulin, glucagon, somatostatin and human pancreatic polypeptide (hPP) by peroxidase antiperoxidase (PAP) method. The pancreas was divided into four regions (principal and secondary islets, exocrine and pancreatic duct regions). In addition, the pancreatic islet regions were further subdivided into three regions (central, mantle and peripheral regions) and the pancreatic duct regions were subdivided into two regions (epithelial and subepithelial regions). Spherical to spindle or occasionally round to oval shaped immunoreactive (IR) cells were demonstrated in the pancreatic islets, exocrine and pancreatic duct. In the principal islet regions, some cells were also detected in the other regions, most of insulin- and somatostatin-IR cells were located in the central regions, and glucagon- and hPP-IR cells were situated in the peripheral regions. In this regions, insulin-IR cells were most predominant cell types and then, glucagon, somatostatin and hPP in that order. In the secondary islet regions, the regional distribution and relative frequency of these four types of endocrine cells were quite similar to those of the principal islets except for cell clusters consisted of hPP-IR cells that were situated in the peripheral to mantle regions. In the pancreatic duct regions, all four major pancreatic endocrine cells were demonstrated in the inter-epithelial cells and/or basal regions of the epithelial linning. In addition, cell clusters composed of numerous insulin-, moderate glucagon- and somatostatin-IR cells of low frequency were also observed in the subepithelial regions of the pancreatic duct. In the exocrine regions, insulin-, glucagon-, somatostatin- and hPP-IR cells were located in the inter-acinus regions with rare, a few, moderate and moderate frequencies, respectively. In conclusion, the regional distribution and relative frequency of four major pancreatic endocrine cells, insulin-, glucagon-, somatostatin- and hPP-IR cells, in the pancreas of the carp showed general patterns which were observed in other stomachless teleost. However, some species- dependent different distributional patterns and/or relative frequencies were also demonstrated.
